Whats Orlistat?

 

Orlistat is a lipase inhibitor used in the treatment of obesity that works by inhibiting fat-metabolizing enzymes. It was originally approved by the FDA in 1999 as the prescription drug Exenical, was approved in 2007 as the over-the-counter medication Alli. Whats the mechanism of action?

 

Orlistat is a drug designed to treat obesity. Its a potent and selective inhibitor of various lipase enzymes responsible for the metabolism of fat. It acts in the gastrointestinal (GI) tract via covalent binding to the serine residues located on the active site of both gastric and pancreatic lipase. When orlistat is taken with food containing fat, it partially inhibits the hydrolysis of triglycerides. This decreases absorption of monoaclglycerides and free fatty acids, contributing to weight maintenance and weight loss.

 

Whats the side effects of the Orlistat?

 

The main side effects of this drug are related to the gastrointestinal tract.

 

Abdominal pain or discomfort:

Oily discharge from the anus

Gas with oily anal discharge

Oily stools

More-frequent bowel movements

Urgent or hard-to-control bowel movements

 

Other possible side effects include:

Headache

Back pain

Upper respiratory infection

 

Orlistat is approved for use in adults 18 and older who have a body mass index (BMI) of 25 or more. Xenical is approved for use in adults with a BMI of 30 or more (obese), and those with a BMI of 27 to 29 (overweight) who have other health risk factors such as high blood pressure or diabetes.
