Retatrutide Phase 3 Results: Weight Reduction Exceeding 30%

The TRIUMPH-1 study evaluated Retatrutide in adults with obesity or overweight and at least one weight-related comorbidity, excluding diabetes.

At week 80, all treatment groups reached both primary and key secondary endpoints with clinically meaningful weight reduction.

Key findings included:

4 mg dose

Average body weight reduction: 19.0%

Average weight loss: 47.2 lbs

9 mg dose

Average body weight reduction: 25.9%

Average weight loss: 64.4 lbs

12 mg dose

Average body weight reduction: 28.3%

Average weight loss: 70.3 lbs

Even more notably, in a predefined extension involving patients with BMI ≥35:

Participants receiving 12 mg treatment through week 104 achieved an average weight reduction of 30.3%.

Historically, weight reduction at this level has typically been associated with bariatric surgery.

In addition to weight reduction, Retatrutide also demonstrated improvements in several cardiometabolic markers, including:

Waist circumference

Non-HDL cholesterol

Triglycerides

Systolic blood pressure

High-sensitivity C-reactive protein (hsCRP)

These results suggest that the therapy may potentially affect broader metabolic health indicators beyond body weight alone.

 

Why Is Retatrutide Receiving So Much Attention?

The answer may lie in its mechanism of action.

Earlier therapies focused on one or two pathways:

Semaglutide

Tirzepatide

GLP-1 + GIP dual agonist
